If we use #setmqcap command to make it effective do we need to restart QMs?

You shouldn't be using setmqcap.

Setmqcap doesn't apply to MQ v6.

All versions of MQ that setmqcap applies to, are out of service at the end of September.

You should not be installing new copies of software earlier than MQ v6.

To address your question a little more directly: What happens when you try it?

Do you have to restart the machine to add more processors?

It will take you exactly two minutes, or less, to try this command on some test server, and find out what happens.
