There is an entire manual dedicated to how to create Queue Manager Clusters.

That seems like a good thing to at least glance at before you start trying to create one.

At least, to me.

how to do it.
The absolute best place to start, when building a cluster is to review the Queue Manager Clusters manual. Chapter 3 specifically takes you through the required steps for creating a simple cluster involving two queue managers: http://publibfp.boulder.ibm.com/epubs/pdf/csqzah06.pdf

...unable to connect: amq4043
AMQ4043 Queue manager not available for connection. Ensure that the queue manager is running. If the queue manager is running on another computer, ensure it is configured to accept remote connections.

Appears to be self-explanatory. Good luck.
