That has so little context it's impossible for us to answer. Also anything with that little context (and that phrasing) probably comes off a certification exam anyway.

Usually you identify things by examining them.

If you are attempting to identify them by using a program, then you write code to identify the things that indicate that a message is a request or a reply. Then you decide if you need to identify which reply goes with which request. Again, you write code to do this.

There's lovely information in the Knowledge Center for MQ about how to write MQ applications using your favorite programming language. This includes information on requests and replies.
