Can we install 6.0.1.0 directly without installing 6.0? or 6.0 is a prerequisite for 6.0.1.0?

No, you can't install just the refresh pack.

Refresh Packs are freely available, original software is not.

Refresh packs contain fix packs and minor functional changes to the base software.
