If I want to connect on an external Database I want to give the connection user in the mqsicreatebroker command with the -u parameter but in which group must the user be defined ? In DB2 Instance ? in broker group =?

It is good enough to add it as a database user. In DB2, the user would be an OS user and in Oracle, it would be an oracle user.
