I found this on IBM website for WF faqs.
Question
What is the maximum number of users?
Answer
For Silvernodes (and comparable machines) 600 users are possible without problems. The maximum number of users recorded is 4000.
On NT boxes we can say a 100 users are OK, 150 are still possible, and 200 would be at the limit (200 users would mean about 800 amqcrsta processes in the system - NT is much less capable (compared to a UNIX system) to handle a real large amount of operating system processes).
I have very little idea about NT/2000. However on AIX 4.3.3 I was running with 1500+ clients with no problem.
Also if you are using Application Server It is recommended to have Queue Manager installed on App box clustred with Remote Queue Manager. This way you have optimized communication. In this scenario your EJB's/Servlet will be opening connection with Local Queue Manager.
