I think its problem of MUSR_MQADMIN accessing the windows file sysstem. Please try the following and let us know what happened.
1. Please check if user ( the one you are using) has access to "c:\program files\IBM\Websphere MQ" directory.
2. Please check if MUSR_MQADMIN is the part of administrator's group. THIS is the most IMPORTANT step.
So after adding please reboot the machine and try creating the queue manager first through command line and then accessing it through MQ Explorer. Hopefully this works.

Tones of thanks Somesh i could create the Local queue manager through exploree now and i can also see the queue manager created from comand prompt at Mq explorer as i add MUSR_MQADMIN to mqm group guided by you
