I want to write a simulator which will send a messages to External system.
The external system will process the message and reply back to response queue. A MDB is listening this response queue and take the messages from the response queueu and process the further steps.

Note: Bothe the request JMS queue and response JMS queue will be maintained by external system.

What is the best approach ?

what is the source of the messages the simulator is going to send? a file? if so, use on of the many file-to-queue programs / samples around (e.g. q programm (MA01 supportpac)). maybe other tools can help too (e.g. rfhutil) if a specific message format is required.
