In cluseter load balancing, does this parameter has to be setup in queue in all qmgr or just in the queue manager where the request comes at the beg? Lets say gateway QMGR.

The attribute at qmgr level has 2 values
(LOCAL | ANY). At queue level it has 3 values (LOCAL | ANY | QMGR).

Pick your choice on where to set it.
The default for the queue is (QMGR).

It only comes into play if the qmgr doing the put/ queue name resolution is hosting the queue.

Prior(In earlier versions), if we had a local cluster queue and a remote cluster queue, messages always went to the local cluster queue. Now it is configurable by using the CLWLUSEQ attribute.

If you don't have local cluster queue (cluster queue hosted by the QM), don't worry about this attribute.

My scenario is like this:

We have a bunch of queue manager ( in cluster) which we call as frondend qmgrs. There is another bunch of qmgrs which we call as backend qmgrs.

Client app connect to the frondend qmgrs and put the message in cluster queue hold physically by the backend qmgrs.

the client ( around 12,000) pick up the qmgrs randomly. Basicaly, our experiences have been that the connection were distributed more or less equally with a few hundred differences. But recently, we came across a situation that the connections are not distributed. Like, some of them haev over 4000, connections whereas some of them haev only around 2000.

So, now we are thinking implementing formal loadbalancing. Formal I mean we were thinking that the app some reasons does that but it was proved untrue.

So, we are considering doing like connecting them to a single qmgrs first which will distributed the load based on the roundrobin fashion.

what is your suggestion? does it work in this scenario or do we need to implement other technology such as BigIP?
