Hi,
Check whether the Option "This person is currently Absent" is checked for the user to whome you are trying to transfer the work item. This can be found in the properties of the user.
1) Look at "Automatically reset ...." option in the same page as well.
2) Look at the Staff Assignment option under the activity tab of Domain, System group, System. (First look at System level, If inherited look at System Group, If inherited look at Domain level and make sure what you have set is correct).
