Hi, we would be changing the 'CERTLABL' property on our queue manager and wondering after changing this value, do we need to do "REFRESH Security" or will it automatically take into affect?

Yes you need to refresh security for it to take effect. And even then, it might not affect running channels...

It should do - any running TLS channels should stop and restart (within the time limit for the platform).

We didn't ran "Refresh Security" command but it worked, the reason might be earlier all client connection was throwing SSL error and after changing the certlabl those connections start connecting with a fresh value of certlabl
