I am working on a .NET API to put messages on MQ Queues. After putting a message successfully do I receive any acknowledgement that the message has been posted successfully to the Queue?

If yes, how is this done in .net.

The object oriented APIs do not give you return codes. They merely fail to throw exceptions.

The regular APIs give you a completion code and return code on every MQ call.
