I need to send my message to a single dynamic destination and also modify the replyToQ & QMgr names from what I was given on my input message.

Should I do this with DestinationList or is there a more efficient way?

You can do this with DeatinationList.

For setting dynamic destination queue,

SET OutputLocalEnvironment.Destination.MQ.DestinationData[1].queueName = <Dynamic queue name from Input message>;
(Note: Output Node should not have queue name and Destination mode should be set to Destination List)

For replyToQ and Qmgr:

SET OutputRoot.MQMD.ReplyToQ = <Queue name from input message>;
SET OutputRoot.MQMD.ReplyToQmgr = (Qmgr name from input message>;

I would use DestinationList to store output queue Name.
You could either use ESQL to change ReplyToQ and ReplyToQmgr fields or specify these values in MQOutput node (under Request tab).
